Decamethylcyclopentasiloxane has not been shown to be genotoxic/mutagenic when tested in a number of short-term in vitro and in vivo assays and did not cause reproductive or developmental toxicity in rats.

Inhalation exposure of rats to 160 ppm Decamethylcyclopentasiloxane for up to 24 months produced adverse effects in the liver (weight changes and hepatocellular hypertrophy) and uterus (increased incidence of endometrial adenocarcinoma, endometrial adenoma, and adenomatous polyps in several animals); however, the results of recent mode-of-action studies are consistent with a uterine tumorigenesis mechanism that is not relevant for humans.
Based on the results of the chronic inhalation study, 160 ppm was determined to be the no-observed-adverse-effect level (NOAEL) and was selected as the point of departure for the derivation of the workplace environmental exposure level (WEEL) value.

This NOAEL was adjusted to account for interindividual variability and residual uncertainty regarding upper respiratory tract changes still occurring at 160 ppm.
The resulting 8-h time-weighted average WEEL value of 10 ppm is expected to provide a significant margin of safety against any potential adverse health effects in workers exposed to airborne Decamethylcyclopentasiloxane.

Following inhalation exposure, exhalation of unchanged Decamethylcyclopentasiloxane and excretion of metabolites with urine are major pathways for clearance in mammals.
Due to this rapid clearance by exhalation, the potential for bioaccumulation of Decamethylcyclopentasiloxane is considered unlikely.

Decamethylcyclopentasiloxane was not DNA reactive or mutagenic in standard in vitro and in vivo test systems.

Decamethylcyclopentasiloxane also did not induce developmental and reproductive toxicity in appropriately performed studies.
In repeated studies in rats with subacute, subchronic and chronic inhalation exposure, mild effects on the respiratory tract typically seen after inhalation of irritating materials, increases in liver weight (28- and 90-day inhalation studies), and a small increase in the incidence of uterine adenocarcinoma (uterine tumor) in female rats (two-year inhalation chronic bioassay) were observed.

The liver effects induced by Decamethylcyclopentasiloxane were consistent with D5 as a weak “phenobarbital-like” inducer of xenobiotic metabolizing enzymes and these effects are considered to be an adaptive response.
Mechanistic studies to elucidate the mode-of-action for uterine tumor induction suggest an interaction of Decamethylcyclopentasiloxane with dopamine signal transduction pathways altering the pituitary control of the estrus cycle.

The resulting estrogen imbalance may cause the small increase in uterine tumor incidence at the highest Decamethylcyclopentasiloxane-exposure concentration over that seen in control rats.
A genotoxic mechanism or a direct endocrine activity of Decamethylcyclopentasiloxane is not supported as a mode-of-action to account for the induction of uterine tumors by the available data.

Production and polymerization of Decamethylcyclopentasiloxane:
Commercially Decamethylcyclopentasiloxane is produced from dimethyldichlorosilane.
Hydrolysis of the dichloride produces a mixture of cyclic dimethylsiloxanes and polydimethylsiloxane.

From this mixture, the cyclic siloxanes including Decamethylcyclopentasiloxane can be removed by distillation.
In the presence of a strong base such as KOH, the polymer/ring mixture is equilibrated, allowing complete conversion to the more volatile cyclic siloxanes:
[(CH3)2SiO]5n → n [(CH3)2SiO]5

where n is a positive integer.
D4 and Decamethylcyclopentasiloxanes are also precursors to the polymer.
The catalyst is again KOH.

History of Decamethylcyclopentasiloxane:
Decamethylcyclopentasiloxane, a type of cyclic siloxane, has a history tied to the development of organosilicon chemistry in the 20th century.
Silicones, as a broader class, were first synthesized in the early 1900s, with significant contributions from British chemist Frederick Kipping, who studied organic silicon compounds.

However, Decamethylcyclopentasiloxane wasn’t until the mid-20th century that cyclic siloxanes, including D5, were produced and became prominent in industrial applications.
D5, with Decamethylcyclopentasiloxane's unique chemical structure, consisting of five silicon-oxygen units in a ring, was found to have exceptional stability and low surface tension, which made it highly desirable for various commercial uses.
By the 1980s and 1990s, Decamethylcyclopentasiloxane became widely used in personal care products, including deodorants, hair sprays, and skin lotions, due to its volatility, smooth texture, and non-greasy feel.

Environmental and health concerns surrounding Decamethylcyclopentasiloxane started to emerge in the early 2000s as researchers began investigating the compound's potential to persist in the environment and bioaccumulate in organisms.
Despite Decamethylcyclopentasiloxane's widespread use, regulatory bodies in different parts of the world, such as the European Chemicals Agency (ECHA) and Environment Canada, began reviewing its safety.

Over the years, studies have led to varied conclusions, with some research suggesting minimal environmental impact, while other investigations raised concerns about Decamethylcyclopentasiloxane’s long-term effects.
The regulatory status of Decamethylcyclopentasiloxane continues to evolve as more research is conducted, balancing its industrial importance with environmental safety.
