The plant Humulus lupulus is known as the raw material of the brewing industry. Hop cones, rich in polyphenolic compounds and acyl phloroglucides, are widely used to preserve beer and to give it a characteristic aroma and flavor. Hop cones have long been used for medicinal purposes. In particular, hop preparations were mainly recommended for the treatment of sleeping disorders. The antioxidative action of hop cones, however, is poorly understood. The aim of our present study was to investigate in vitro changes in human blood platelets induced by peroxynitrite (ONOO(-), the compound of particular importance for vascular thrombosis and inflammatory process) in the presence of hop cone extract (Humulus lupulus). The antioxidative action of the extract was also compared with the properties of a well-characterized antioxidative commercial monomeric polyphenol, resveratrol (3,4',5-trihydroxystilbene) in a model system in vitro. Various biomarkers of oxidative/nitrative stress, such as carbonyl groups, 3-nitrotyrosi
ne and thiobarbituric acid reactive substances (TBARS) were estimated. The 3-nitrotyrosine formation and carbonyl group generation was assessed by the use of a competition ELISA test and ELISA test, respectively. Tested plant extract (12.5-50 µg/ml), like resveratrol, significantly inhibited protein carbonylation and nitration in the blood platelets treated with ONOO(-) (0.1 mM). The extract from hop cones, like resveratrol, also caused a distinct reduction of platelet lipid peroxidation induced by ONOO(-). The present results indicate that the hope cone extract has in vitro protective effects against ONOO(-), such as induced oxidative/nitrative damage to the human platelet proteins and lipids. However, in comparative studies the extract was not found to be a more effective antioxidant than the solution of pure resveratrol.


Hops (Humulus lupulus) are cultivated for their use in beer making; the flowers of the female plant are used to impart the bitter flavor and pungent aroma to beer. Ingestion of both fresh and spent hops has been associated with the development of malignant hyperthermia in dogs.35 Breeds that may appear to be particularly susceptible to this problem include greyhounds and golden retrievers. Treatment is directed toward basic decontamination of the gastrointestinal tract if the exposure is recent, and controlling the high temperatures in clinically affected animals (e.g., IV fluids, various cooling measures). If available, dantrolene, a skeletal muscle relaxant that has been used in humans to manage malignant hyperthermia, may be used at an initial dose of 2-3 mg/kg IV or 3.5 mg/kg PO followed by 3.5 mg/kg every 12 hours as needed.

 


Hop (Humulus lupulus L., family Cannabaceae) is a dioecious perennial climbing plant grown commercially worldwide for its use in the pharmaceutical and brewing industries. Traditionally the annual stems (vines/bines) are pruned continuously during the season to promote growth of selected vines and to control disease. After harvest, shoots are trimmed to soil level for the winter; in spring, new shoots arise from the rootstock.

 

Although no in vitro studies, directly related to the sedative activity of hop strobiles have been published, several studies in animals have been carried out to investigate the neuropharmacological properties of Humulus lupulus, traditionally used in the treatment of different CNS disorders such as insomnia, excitability and restlessness.

 

Attached is the Draft Report of 6 Humulus lupulus (hops)-derived ingredients as used in cosmetics. [HumLup092016Rep] These ingredients are derived from parts of the Humulus lupulus (hops) plant. The strobiles (also called cones) of Humulus lupulus are known for their use in beer brewing.

 


The Panel should note the sensitization data on people who work in the cultivation of the Humulus lupulus (hops) plant and in the beer industries as well as the data on the potential estrogenic effects.

 


All of these ingredients are derived from part or all of the Humulus lupulus (commonly called hops) plant. This plant is generally known as an ingredient in the brewing of beer.2,3 The terms "cone" and "strobile" are synonymous and refer to the structures formed by the female Humulus lupulus (hops) plant after the flowers have bloomed, whether or not they have been fertilized. Thus, "cone oil" is the same as "strobile oil". Both "cone" and "strobile" are used in the literature. Also, "hops" is used commonly for the plant name as well as for the harvested strobiles; in the literature, it is not always clear if authors are referring to the whole plant or just the strobiles. In this report, it is assumed that the authors are referring to just the strobiles unless otherwise indicated. The Cosmetic Ingredient Review (CIR) Expert Panel (Panel) has reviewed related ingredients. These reviews may provide additional relevant information about Humulus lupulus (hops)-derived ingredients. The Panel reviewed the safety of phytosterols (plant-derived sterols that are likely constituents of most of the Humulus lupulus (hops)-derived ingredients) in 2013 and concluded that the phytosterols are safe as used.

 

The Panel also reviewed citrus peel oils, which are essential oils like Humulus Lupulus (Hops) Cone Oil, in 2014 and concluded that these ingredients are safe for use in cosmetic products when finished products, excluding rinse-off products, do not contain more than 0.0015% (15 ppm) 5-methoxypsoralen (5- MOP), and when formulated to be non-sensitizing and non-irritating.5 Humulus lupulus (hops) strobile is an ingredient in food (most commonly in beer) and most of the other parts of this plant (shoots, leaves, flowers, seeds, rhizomes, and essential oils) are edible. 6,7 Also, the U.S. Food and Drug Administration (FDA) determined that essential oils, oleoresins (solvent-free), and natural extractives (including distillates) of Humulus lupulus L. (hops) are generally recognized as safe (GRAS) for human consumption.[21CFR182.20] Consumption of these Humulus lupulus (hops)-derived ingredients would result in much larger systemic exposures than what is expected from use in cosmetic products, even if there was 100% absorption. Thus, the systemic toxicity potential of Humulus lupulus (hops)- derived ingredients will not be the focus of this safety assessment (although toxicity information may be included here). The primary focus of the safety assessment is the review of safety based on local (dermal) effects. Botanical cosmetic ingredients, such as Humulus lupulus (hops)-derived ingredients, may contain hundreds of constituents, some of which could have the potential to cause toxic effects. For example, sesquiterpene lactones present in Humulus lupulus (hops) plant may cause Type IV allergic reactions (cell-mediated, delayed-type hypersensitivity) and toxicity when present in sufficient amounts, and β-myrcene may be a dermal irritant and sensitizer. 8,9 In this assessment, CIR is reviewing information available to evaluate the potential toxicity of each Humulus lupulus (hops)-derived ingredient as a whole, complex substance. Except for specific constituents of concern, CIR is not reviewing information that may be available to assess the potential toxicity of the individual constituents derived from Humulus lupulus (hops). The names of the ingredients in this report are written in accordance with the Dictionary, as listed above, capitalized without italics and without abbreviations. When referring to the plant from which these ingredients are derived, the standard taxonomic practice of using italics will be followed (e.g., Humulus lupulus).

 

In general, when the strobiles are harvested from Humulus lupulus (hops) bines, the strobiles are immediately dried by forced hot air, and are often pressed into dense cylindrical pellets, 5 to 8 mm in diameter and up to 25 mm long.16 The pelletization reduces the overall surface area and therefore reduces the rate of chemical oxidation/degradation, and provides a more compact product for shipping. Not all suppliers dry the strobiles; the strobiles may be harvested fresh as whole, wet cones at the farm, and shipped for immediate use within 36 h of harvest.10Sometimes, hops are treated with sulfur dioxide to improve the color and prevent change of active constituents.

 

In general, oils are extracted from Humulus lupulus (hops) by steam distillation at 100˚C.22 Methylene chloride is the most common solvent used for the extraction of Humulus lupulus (hops) for beer brewing; hexane and methanol are also employed.23 Typically, at least 95% of the available α-acids (source of the bitter flavorings) can be extracted from fresh Humulus lupulus (hops) strobiles. Methanol is the most efficient solvent for the extraction of α-acids (approximately 25%), followed by methylene chloride (approximately 20%) and hexane (approximately 18%). Supercritical fluid extraction (SFE) with carbon dioxide is also used to collect the extract for beer brewing.24-27 Another modern method is pressurized fluid extraction (PFE), which is employed for extracting different polyphenols.28-32 PFE has been developed to extract bitter acids.

 


According to the VCRP survey data received in 2016, Humulus Lupulus (Hops) Extract was reported to be used in 362 formulations, including 299 leave-on formulations and 60 rinse-off formulations (Table 11).56 Humulus Lupulus (Hops) Cone Extract and Humulus Lupulus (Hops) Strobile were reported to be used in 4 and 7 cosmetic formulations, respectively. The results of the concentration of use survey conducted by the Council in 2016 indicate that Humulus Lupulus (Hops) Extract has the highest reported maximum concentration of use; it is used at up to 0.6% in face and neck products.57 The maximum concentration of use for Humulus Lupulus (Hops) Cone Extract and Humulus Lupulus (Hops) Flower Extract are reported to be 0.00055% (face powders) and 0.001% (hair conditioners and other hair preparations), respectively. In some cases, reports of uses were received in the VCRP, but no concentration of use data are available. For example, Humulus Lupulus (Hops) Strobile was reported to be used in 7 formulations, but no use concentration data were available. In some cases, no uses were reported in the VCRP, but concentration of use data were received from industry; Humulus Lupulus (Hops) Flower Extract had no reported uses in the VCRP, but use concentrations in two hair product categories and bath soaps and detergents were provided in the industry survey. Therefore, it should be presumed there is at least one use in every category for which a concentration is reported. The ingredients not in use according to the VCRP and the industry survey are Humulus Lupulus (Hops) Cone Oil and Humulus Lupulus (Hops) Stem Extract. Humulus Lupulus (Hops) Extract (up to 0.13%), Humulus Lupulus (Hops) Cone Extract, and Humulus Lupulusm (Hops) Strobile are reported to be used in formulations that are used around the eyes. Humulus Lupulus (Hops) Extract (up to 0.084%), Humulus Lupulus (Hops) Cone Extract (no concentration of use reported), and Humulus Lupulus (Hops) FlowermExtract (up to 0.000055%) are reported to be used in formulations that come in contact with mucus membranes (e.g., bathü soaps and detergents, bubble baths). Humulus lupulus (Hops)-derived ingredients are used in cosmetic sprays and could possibly be inhaled; for example, Humulus Lupulus (Hops) Extract is reported to be used at up to 0.0002% in hair sprays. In practice, 95% to 99% of the droplets/particles released from cosmetic sprays have aerodynamic equivalent diameters >10 µm, with propellant sprays yielding a greater fraction of droplets/particles <10 µm compared with pump sprays.58,59 Therefore, most droplets/particles incidentally inhaled from cosmetic sprays would be deposited in the nasopharyngeal and thoracic regions of the respiratory tract and would not be respirable (i.e., they would not enter the lungs) to any appreciable amount.60,61 Humulus Lupulus (Hops) Cone Extract is used in face powders at up to 0.00055%.

 

Humulus lupulus is a flowering plant, also known as common hops, it is native to temperate regions of Northern hemisphere. The plant extract and the strobiles of cumulus lupulus are particularly useful in cosmetics. Hops has been used for beer making for years and cultivated for the same reason. It contains resinous bitter compound humulone, lupulone, xanthium, humulene, tannins, polyphenols, protein, waxes, steroids, bitter acids called alpha acids and beta acids, cellulose, pectins, etc. The extract is a golden yellow colored liquid.

Humulus lupulus extract is mainly antimicrobial in nature. Medicinally it is used to treat various skin and hair ailments. For cosmetics, this property has a dual purpose. First, it also helps preserve formulation free from any microbial contamination. Second, it helps skin get rid of harmful bacteria that can cause acne or body odor. When the extract is applied on skin it forms a layer on the skin that protects the skin from outer environment, it preserves skin's moisture and makes it available for skin cells. So, the skin remains moisturized and soft. It also helps remove the dead skin layer from the epidermis and cleanses the skin. It is used in skincare and haircare formulations.

Humulus lupulus L. (hops) is a popular botanical dietary supplement used by women as a sleep aid and for postmenopausal symptom relief. In addition to its efficacy for menopausal symptoms, hops can also modulate the chemical estrogen carcinogenesis pathway and potentially protect women from breast cancer.

Humulus lupulus, Kendirgiller familyasındandır. Anayurdu Kuzey Amerika ve Avrasya olan, belli mevsimde kısmen ölen ama çokyıllık olan tırmanıcı bitkidir. Bira endüstrisi için Avrupada geniş ölçüde tarımı yapılan bitki, ülkemizde aynı amaçla Bilecik ve Bursa yörelerinde üretilmekte, ayrıca Kuzey Anadoluda kendi kendine doğada yetişen şerbetçiotu örnekleri seyrek de olsa görülmektedir. Yetişmesi için ılıman iklim ve zengin toprağa: sarılması için yüksek direk ve aralarına gerilmiş tellere gereksinen şerbetçiotu, 7,5 mye kadar uzayabilir. Kırmızımsı yeşil renkli ince ve sert gövdesi, neyi bulursa ona sarılıp uzar: ama yaz sonunda yapraklarıyla birlikte ölür. Ertesi yıl aynı kökten yeni sürgünleri çıkar. Asmanınkileri andıran yaprakları uzun saplı, 3-5 parçalı, iri, karşılıklı dizili, kenarları dişli, normal yeşil renkli ve kalp biçiminde olur. Yaz sonunda açan kozalakçığa benzeyen sarımsı yeşil renkli dişi çiçek tomurcuklan olgunlaşınca irileşir ve kâğıdımsı bürgülere dönüşür. Bitkinin erkek çiçekleri ayrı bitkide açar. Şerbetçiotu bitkisi tohumuyla, kökleri bölünerek ya da genç piçleri ilkbaharda sökülüp ayrı yere dikilerek çoğaltılır. Şerbetçiotu bitkisi uçucu yağ, acı maddeler (humilon, lupulon), reçine, tanen ve mum içerir. Dişi çiçekleri strobili lupuli adlı bir maddeyi oluşturur. Bu madde en az 1200 yıldan beri bira yapımında kullanılmaktadır. Şerbetçiotunun genç sürgünleri bazı ülkelerde soyulup kuşkonmaz gibi pişirilerek yenir.

Bu etkileri sağlamak üzere, olgunlaşmamış çiçek kozalakçıkları yaz sonu ile sonbahar başında toplanıp gölgeli, havadar bir yerde özenle kurutulur. Böyle kurutulmuş çiçeklerden 1 tatlı kaşığının üzerine 1 bardak kaynar su dökülerek 10-15 dakika demlendirilip infüzyon hazırlanır. Bu infüzyondan günde bir bardak içilir.
• Şerbetçiotu, merkezi sinir sistemi üzerindeki yatıştırıcı etkisiyle aynı zamanda iyi bir uyku ilacı etkisi yapar.
• Bazı kişilerde şehveti yok edici (anafrodizyak) etki gösterir.

Methanol ve n-Hexan ekstraktlarının patojenlere karşı, 0.1, 0.5, 1 ve 2 mg/ml dozlarının Miselyum Gelişimi (MG), Miselyum Gelişim Engelleme (MGE) etkinlik düzeyleri belirlenmiş ve letal doz (LD₅₀) hesaplanmıştır. LD₅₀ hexan ve methanol ekstratları için Sc için; sırasıyla 0.021 ile 0.193 mg/ml, Fol için; sırasıyla 0.616 ile 0.273 mg/ml olarak hesaplanmıştır. Çalışma sonucunda; H. lupulus çiçeklerinden elde edilen Methanol ve n-Hexan ekstraktlarının in vitro şartlarda Sclerotinia sclerotiorum ve Fusarium oxysporum f. sp. lycopersici üzerine fungusidal aktivitelerinin olduğu belirlenmiştir.

Humulus lupulus, ortak şerbetçiotu olarak da bilinen çiçekli bir bitkidir, Kuzey yarımkürenin ılıman bölgelerine özgüdür. Bitki özütü ve kümülüs lupulus strobilleri özellikle kozmetikte yararlıdır.
